HDX-MS (hydrogen-deuterium exchange mass spectrometry) technology is a mass spectrometry technique for studying the spatial conformation of proteins. The principle of this technique is to immerse protein molecules in a deuterium-containing aqueous solution and exchange amide hydrogens of the protein molecule with deuterium atoms from the solution at different rates. This process depends on the protein configuration to a certain extent, and the hydrogen-deuterium exchange rate of protein fragments is determined by mass spectrometry so as to obtain information regarding the spatial structure of the protein as well as interpret the degree of activity of different sites.
